Handling procedure
Open vial according to enclosed instructions.  Prepare biphasic slants by adding 2-3 mL of #1928 broth over a #1921 slant.  
Under anaerobic conditions, withdraw 0.5 mL of #1928 broth from the biphasic slant and rehydrate the entire vial contents.
Aseptically transfer this aliquot back into the biphasic slant tube. Inoculate #1921 agar slants, broths and/or plates with 0.2 mL each of the cell suspension. Additional #1928 broth biphasic slant tubes may be inoculated with 0.2 mL each. An aerobic agar blood plate may also be streaked to check for purity.
Incubate the #1921 tubes and plates under anaerobic conditions at 37°C. Incubate agar blood plate aerobically at 37°C.
Within approximately seven days, growth should be evident by off white growth on the bottom of the biphasic slant.  No growth should occur on the blood agar plate incubated aerobically.


ANAEROBIC CONDITIONS:
Anaerobic conditions for transfer may be obtained by either of the following:

Use of an anaerobic gas chamber;
Placement of test tubes under a gassing cannula system hooked to anaerobic gas.
Anaerobic conditions for incubation may be obtained by any of the following:

Loosen screw-caps on test tubes in anaerobic chamber;
Loosen screw-caps on test tubes in an activated anaerobic gas pack jar,
Use of sterile butyl rubber stoppers on test tubes so that an anaerobic gas headspace is retained.
Handling notes
NAM is required for growth.
To achieve optimal growth, grow this item biphasically.  Weak growth in broth and on agar can only be achieved after extended incubation.
Always use freshly prepared pre-reduced media or pre-reduced media that has been previously prepared but stored under anaerobic conditions.
