Cytogenetic Analysis: Chromosome Frequency Distribution 50Cells: 2n = 44. This is a pseudodiploid line with the tetraploidy occurring at4%. The karyotype is 44,XY,-6,-15,6q+,15q+ in a majority of cells analyzed. Themarkers 6q+ and 15q+ occurred in most cells. An occasional monosomic ortrisomic condition for a normal chromosome was also detected.

Comments: Theparent line of BHK-21(C-13) was derived from the kidneys of five unsexed,1-day-old hamsters in March, 1961, by I.A. Macpherson and M.G.P. Stoker.
Following 84 days of continuouscultivation, interrupted only by an 8-day preservation by freezing, clone 13was initiated by single-cell isolation.
This line has been used as a host fortransformation with expression vectors containing selectable and amplifiablemarker DNAs (e.g., Factor VIII).
The World Organization for Animal Health(OIE) uses the cells for routine diagnosis of rabies. (see:http://www.oie.int/Eng/Normes/Mmanual/A_00044.htm).
Propagation: Eagle's Minimum Essential Medium, Catalog No. 30-2003. To makethe complete growth medium, add the following components to the base medium:fetal bovine serum to a final concentration of 10%.
Temperature: 37.0°C
Subculturing: Protocol: Remove medium, and rinse with 0.25% trypsin, 0.03% EDTAsolution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTAsolution. Allow the flask to sit at room temperature (or at 37C) until thecells detach. Add fresh culture medium, aspirate and dispense into new cultureflasks.
Subcultivation Ratio: A subcultivationratio of 1:2 to 1:10 is recommended
