Description: gWIZ vectors representa new series of plasmids that have beenengineered to produce the highest levelsof transgene expression in a wide range ofmammalian cells and tissues. It contains aproprietarily modified promoter followedby the intron A from the humancytomegalovirus (CMV) immediate earlygene and a high-efficiency artificialtranscription terminator. The expressionvector is constructed in the context of aplasmid backbone extensively modified toachieve the enhanced levels of trangeneexpression in mammalian cells as well ashigh efficiency of plasmid production inE. coli.Components: 25 µg gWIZ GFP plasmid in25 µl sterile TE buffer.Storage: Store at -20oC.Comments: gWIZ is suitable for in vitro and in vivogene expression studies and applications.Use Kanamycin as selection to grow theplasmid in E. coli.INTRODUCTIONThe CMV immediate early gene (IE) promoter/enhancer isthe most widely used constitutive promoter for expressinghigh levels of trangene product in many mammalian cellsand tissues. However, not all CMV IE genepromoter/enhancer-based expression vectors are createdequal. Depending on the actual CMV IE gene sequencesused and the context of the plasmid backbone upon whichthe expression cassette is constructed, the expression levelscan vary as much as two orders of magnitude. The CMV IEpromoter sequences contained in the gWIZ vectors havebeen systemically analyzed and modified. Themodifications include removing the sequences that areredundant and deleterious to the high levels of expressionwhile retaining those sequences that are of hightranscriptional potency. After coupling the modifiedpromoter with a high-efficiency synthetic transcriptionalterminator, the whole expression cassette is finallyconstructed on a plasmid backbone that has also beenstreamlined and modified to accommodate the high levels ofexpression in mammalian cells as well as high yield ofplasmid production in E. coli. The resulting plasmid, gWIZexpression vector, is capable of fully unleashing thepotential of the CMV promoter and giving the highest levelsof expression possible both in vitro and in vivo.USAGE• For extremely high levels of transgene expression inmammalian cells and tissues• Can be used with GenePORTER 2 (Cat. # T202007 orT202015) to transfect a wide variety of mammalian cellsand tissuesDETECTION OF THE EXPRESSED GENEGreen Fluorescent Protein: For transfections conductedwith plasmids encoding GFP, the detection can be done byepifluorescence or confocal microscopy. The GFP producedhas an excitation peak at 470-480 nm and emission peak at510 nm. Expression level of GFP can also be monitored byfluorescence-activated cell sorter analysis as described by L.Cheng et al. * The gWIZ-GFP plasmid contains a modifiedand much brighter version of green fluorescent protein gene.*Cheng, L. et al.
