Description

Murine Bladder Carcinoma Cell Line Transfected With Luciferase (MB49-Luc-mCh-Puro) are parental MB49 tumor cells transfected with a Luc expression plasmid using Lipofectamine for luciferase expression. Parental MB49, a murine transitional cell bladder carcinoma line, forms tumors when injected into mouse bladders, either orthotopically or subcutaneously. This cell line is suitable for use in studies concerning bladder carcinoma requiring in vivo tumor imaging.

Propagation Requirements

Use of PriCoatT25 Flasksor Applied Cell Extracellular Matrixs required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels unless otherwise specified in the Propagation Requirements below.


The base medium for this cell line is Prigrow III medium available at BioVector, Cat. No. BioVector-TM003. To make the complete growth medium, add the following components to the base medium: heat-inactivated fetal bovine serum (BioVector-TM999) to a final concentration of 10%, Penicillin/Streptomycin Solution (BioVector-G255) to a final concentration of 1%, NEAA to a final concentration of 1X, sodium pyruvate (BioVector-TM057) to a final concentration of 1 mM, and L-glutamine (BioVector-G275) to a final concentration of 2 mM.


Carbon dioxide (CO2): 5%, Temperature: 37.0°C.

【Complete Growth Medium完全培养基】 The base medium for this cell line is DMEM/RPMI-1640 Medium. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
【Subculturing传代方法】
Volumes are given for a 75 cm2 flask. Increase or decrease the amount of dissociation medium needed proportionally for culture vessels of other sizes.
Remove and discard culture medium.
Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor.
Add 2.0 to 3.0 mL of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37°C to facilitate dispersal.
Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gently pipetting.
Add appropriate aliquots of the cell suspension to new culture vessels.
Incubate cultures at 37°C.
Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:6 is recommended
Medium Renewal: 2 to 3 times per week
