established in 1994 from the peripheral blood of a 63-year-old woman with secondary acute myelomonocytic leukemia (sAML M4) 25 years after onset of myelodysplastic syndrom (MDS) and 2 years after chemotherapy with Ara-C; cells were described in the literature to carry JAK2 and TP53 mutations and a deletion of the 5q31 AML/MDS region effected by a t(5;11) translocation and to be constitutively cytokine-dependent

80% alpha-MEM (with ribo- and deoxyribonucleosides) + 20% h.i. FBS + 10 ng/ml GM-CSF or 10% vol conditioned medium of cell line 5637 (NTCC930035)
Subculture:
seed out at ca. 0.8 x 106 cells/ml in 24-well plates; maintain at ca. 0.5-1.0 x 106 cells/ml; split culture 1:2 once or twice a week; after thawing cells may need some days to recover and a certain amount of cell debris may be allways visible; CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus, the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines.
