Description / 描述:
The BioVector® pAmpho plasmid is a retroviral envelope expression vector designed for the production of amphotropic retroviruses. It expresses the 4070A amphotropic envelope protein under the control of the CMV immediate-early promoter . The amphotropic envelope protein enables viral particles to infect a wide range of mammalian cells by interacting with the RAM-1 receptor on the host cell surface . This plasmid is commonly used in conjunction with retroviral packaging cell lines and helper constructs to generate replication-incompetent retroviruses for gene delivery .

Storage / 存储:

  • Plasmid is sent as a bacterial stab in agar or as purified DNA. Upon receipt, streak onto a plate with BioVector® Ampicillin (100 μg/mL), grow a single colony, and prepare a glycerol stock for long-term storage at -80°C.

  • 质粒以细菌穿刺菌或纯化DNA形式提供。收到后,应在含 BioVector® 氨苄青霉素 (100 μg/mL) 的平板上划线,挑取单克隆,并制备甘油菌于-80°C长期保存。

Plasmid Reconstitution Protocol / 质粒溶解复配方案:

  1. Centrifuge at 5,000×g for 5 min. / 5,000×g离心5分钟。

  2. Carefully open the tube and add 20 μl of sterile water to dissolve the DNA. / 小心打开管盖,加入20 μl无菌水溶解DNA。

  3. Close the tube and incubate for 10 minutes at room temperature. / 盖紧管盖,室温孵育10分钟。

  4. Briefly vortex and spin down. / 短暂涡旋并离心收集。

  5. Store the plasmid at -20°C. / 于-20°C保存质粒。

  6. It is recommended to transform and extract fresh plasmid before use. / 建议使用前进行转化并提取新鲜质粒。

General Plasmid Transformation Protocol / 通用质粒转化方案:

  1. Thaw 100 μl competent cells on ice for 10 min, add 2 μl plasmid, incubate on ice for 30 min, heat-shock at 42°C for 60 sec, then incubate on ice for 2 min. / 取100 μl感受态细胞冰浴10分钟,加入2 μl质粒,冰浴30分钟,42°C热激60秒,再冰浴2分钟。

  2. Add 900 μl antibiotic-free LB medium, incubate at 37°C for 45 min with shaking at 180 rpm. / 加入900 μl无抗生素LB培养基,37°C 180 rpm振荡培养45分钟。

  3. Centrifuge at 6,000 rpm for 5 min, resuspend pellet in 100 μl supernatant, spread onto LB plate with appropriate antibiotic. / 6,000 rpm离心5分钟,用100 μl上清重悬沉淀,涂布于含相应抗生素的LB平板。

  4. Incubate plate inverted at 37°C for 14 hours. / 倒置平板,37°C培养14小时。

  5. Pick single colony into LB medium with antibiotic, incubate at 220 rpm for 14 hours, extract plasmid. / 挑取单克隆至含抗生素的LB培养基,220 rpm培养14小时,提取质粒。
