This specific mutation is the most prevalent mechanism globally (often exceeding 70%) for acquired resistance to clarithromycin in H. pylori. The A2143G mutation prevents clarithromycin from binding effectively to the 50S ribosomal subunit, thereby neutralizing its ability to inhibit bacterial protein synthesis. 2. Culture Conditions

• Media: BioVector® Columbia Blood Agar (CBA) or Wilkins-Chalgren Agar, supplemented with 7% - 10% defibrinated sheep blood.

• Environment:Microaerophilic conditions ($5\% \ O_2, 10\% \ CO_2, 85\% \ N_2$).

• Temperature: 37 degrees Celsius.

• Humidity: High humidity is essential (use of a water tray is recommended).

• Growth Period: Small, translucent colonies appear within 3 to 5 days.
