一、 背景与来源说明
MDOK(Madin-Darby Ovine Kidney)细胞株衍生自正常绵羊(Ovis aries)的肾脏组织,由 Madin 和 Darby 建立。该细胞系保持着完好的上皮细胞形态与贴壁生长的特性,是兽医病毒学、绵羊及反刍动物相关病毒(如绵羊肺炎病毒、蓝舌病病毒 BTV、伪狂犬病病毒、绵羊痘病毒等)的分离、扩增、滴定测定(TCID50 / Plaque Assay)以及疫苗生产的标准质控细胞模型。
二、 细胞生物学特征
组织来源:绵羊(Ovis aries)肾脏上皮组织(Kidney Epithelium)
细胞形态:贴壁生长,呈典型铺砖状上皮细胞样(Epithelial-like)形态
病毒敏感性(Viral Susceptibility):
高度敏感:蓝舌病病毒(Bluetongue Virus, BTV)、绵羊进行性肺炎病毒(Maedi-Visna Virus)、小反刍兽疫病毒(PPRV)、绵羊痘病毒(Sheeppox Virus)。
部分敏感:口蹄疫病毒(FMDV)、牛病毒性腹泻病毒(BVDV)、伪狂犬病病毒(PRV)。
核型与代数:倍体及代数稳定性高,常规实验推荐在 P10 – P30 代内进行。
三、 培养条件与传代指南
1. 完全培养基配方
基础培养基:MEM(Minimum Essential Medium)或 DMEM (High Glucose)
血清添加:10% 优质胎牛血清(FBS,建议灭活并经无外源病毒/无支原体检测)
关键添加物:
非必需氨基酸(NEAA):1×
丙酮酸钠(Sodium Pyruvate):1 mM
L-谷氨酰胺(L-Glutamine):2 mM
双抗(Penicillin-Streptomycin):100 U/mL 青霉素 + 100 ug/mL 链霉素
2. 培养环境与传代规范
培养环境:37°C,5% CO2,饱和湿度培养箱。
传代比例:1:3 至 1:6 传代,当细胞密度达 80% – 90% 时进行传代(避免过度密合导致细胞老化脱落)。
消化操作:使用 0.25% Trypsin - 0.02% EDTA,37°C 消化 2–3 分钟,待显微镜下细胞变圆脱落后,加入含血清培养基终止消化,轻柔吹打分散成单细胞悬液。

1. Background & Context
The MDOK (Madin-Darby Ovine Kidney) cell line was established from normal kidney tissue of a sheep (Ovis aries) by Madin and Darby. MDOK cells display characteristic epithelial morphology and grow as a confluent monolayer. It serves as an essential host system in veterinary virology for the isolation, propagation, plaque assay titration, and vaccine manufacturing quality control of various ovine and ruminant viruses, including Bluetongue Virus (BTV), Maedi-Visna Virus, and Peste des Petits Ruminants Virus (PPRV).
2. Biological Characteristics
Organism & Tissue:Ovis aries (Sheep), kidney epithelium.
Growth Properties: Adherent, cobblestone-like epithelial monolayer.
Viral Susceptibility Spectrum:
Highly Susceptible: Bluetongue Virus (BTV), Maedi-Visna Virus, Peste des Petits Ruminants Virus (PPRV), Sheeppox Virus.
Permissive/Moderately Susceptible: Foot-and-Mouth Disease Virus (FMDV), Bovine Viral Diarrhea Virus (BVDV), Pseudorabies Virus (PRV).

3. Culture & Handling Protocols
3.1 Complete Growth Medium Formulations
Basal Medium: MEM (Minimum Essential Medium) or DMEM (High Glucose).
Serum Supplement: 10% Fetal Bovine Serum (FBS, qualified/virus-screened).
Supplements:
Non-Essential Amino Acids (NEAA): 1x
Sodium Pyruvate: 1 mM
L-Glutamine: 2 mM
Antibiotics: 100 U/mL Penicillin + 100 ug/mL Streptomycin
3.2 Incubation & Subculture Parameters
Incubation Conditions: 37°C, 5% CO2, humidified atmosphere.
Subculture Ratio & Frequency: 1:3 to 1:6 split ratio, passaged every 3–4 days at 80% – 90% confluency.
Detachment: Treat with 0.25% Trypsin - 0.02% EDTA at 37°C for 2–3 minutes. Inactivate with complete serum-containing medium and gently dissociate into a single-cell suspension.
