三、 培养条件与传代指南
1. 完全培养基配方
基础培养基:RPMI-1640 Medium
血清添加:10% – 15% 优质胎牛血清(FBS)
添加剂:
L-谷氨酰胺(L-Glutamine):2 mM
丙酮酸钠(Sodium Pyruvate):1 mM
双抗(Penicillin-Streptomycin):100 U/mL 青霉素 + 100 ug/mL 链霉素
2. 培养环境与传代规范
培养环境:37°C,5% CO2,饱和湿度。
维持密度:建议将细胞密度维持在 $2 \times 10^5 \text{ cells/mL} - 1.2 \times 10^6 \text{ cells/mL}$ 之间。当密度超过 $1.5 \times 10^6 \text{ cells/mL}$ 时会导致细胞营养耗竭并出现凋亡。
传代操作:悬浮细胞无需胰酶(Trypsin)消化。传代时只需轻柔吹打均匀,按 1:2 至 1:4 的比例半换液或离心(1000 rpm, 5 分钟)重悬接种于新鲜培养基中。
