一、 背景与来源说明 / Background & Origin

MUTU-III(常写作 MUTU 3Mutu-3)是一种来源于人类地方性 Epstein-Barr 病毒(EBV)阳性伯基特淋巴瘤(Burkitt Lymphoma, BL)的 B 淋巴细胞株

MUTU 系列细胞株建立自同一名携带 $t(8;14)$ 染色体易位的男性 BL 患者活检组织(MUTU-BL)的连续传代克隆。其中,MUTU-III 属于 EBV 潜伏期 III 型(Latency Type III)表现型克隆,高表达全套 EBV 潜伏期蛋白(包括 EBNA1、EBNA2、EBNA3 系列、EBNA-LP 以及 LMP1、LMP2 等)及相关微 RNA(ebv-miR-BHFR1-2-3p)。与之相对的姐妹克隆 MUTU-I 保持潜伏期 I 型(仅表达 EBNA1)。该细胞株是研究 EBV 潜伏期类型转换、致瘤机制、EBV 编码蛋白(如 LMP1/EBNA2)信号通路及 B 细胞淋巴瘤免疫逃逸的经典细胞模型。

三、 培养条件与传代指南 / Culture & Handling Protocols

1. 推荐培养基 / Complete Growth Medium

  • 基础培养基:RPMI-1640

  • 添加成分:10% 至 20% 灭活胎牛血清(FBS) + 1% 双抗(Penicillin-Streptomycin,可选)

  • 培养环境:37 摄氏度,5% $CO_2$,极高湿度培养箱

2. 复苏与传代操作 / Thawing & Subculture

  1. 复苏:将冻存管快速置于 37 摄氏度水浴中摇晃融化(约 1-2 分钟)。吸出细胞悬液加至含 8-10 mL 预热完整培养基的离心管中,125 x g 离心 5 分钟。弃上清,用预热培养基重悬,接种至 T25 培养瓶,接种密度建议在 $0.5 \times 10^6\text{ cells/mL}$ 以上。

  2. 传代与维持

    • 维持细胞密度在 $0.3 - 0.5 \times 10^6\text{ cells/mL}$

    • 每 2 至 3 天按 1:2 或 1:3 比例补加新培养基或稀释传代。

    • 最大生长密度一般不超过 $1.0 \times 10^6\text{ cells/mL}$

3. 冻存保存 / Cryopreservation

  • 冻存液配方:70% RPMI-1640 + 20% FBS + 10% DMSO

  • 冻存密度$3 - 5 \times 10^6\text{ cells/vial}$,采用程序降温冻存并长期保存于液氮(-196 摄氏度)。

BioVector® MUTU-III Cell Line

1. Background and Origin

MUTU-III (also designated as MUTU 3 or Mutu-3) is a human B-cell line derived from an endemic Epstein-Barr virus (EBV)-positive Burkitt lymphoma (BL).

The MUTU cell line family was established by serial passaging of the parental clone MUTU-BL, isolated from a biopsy of a male patient carrying the classic $t(8;14)$ translocation.MUTU-III represents the EBV Latency Type III phenotype, characterized by high expression of the full spectrum of latent viral proteins (including EBNA1, EBNA2, EBNA3s, EBNA-LP, and LMP1/LMP2) and viral microRNAs.Its sister clone, MUTU-I, maintains a restricted Type I latency (expressing only EBNA1). MUTU-III serves as an essential cell model for studying EBV latency switching, viral oncogenesis, LMP1/EBNA2 pathway signaling, and immune evasion mechanisms.

Thawing and Subculture Protocol

  1. Thawing: Thaw the frozen vial rapidly in a 37 degrees Celsius water bath. Transfer contents into 8–10 mL pre-warmed complete medium and centrifuge at 125 x g for 5 minutes. Resuspend in fresh medium and seed at $\ge 0.5 \times 10^6\text{ cells/mL}$.

  2. Subculturing:

    • Maintain working cell density between $0.3 - 0.5 \times 10^6\text{ cells/mL}$.

    • Split cells 1:2 to 1:3 every 2–3 days by adding fresh medium.

    • Do not allow density to exceed $1.0 \times 10^6\text{ cells/mL}$.

Cryopreservation

  • Freezing Medium: 70% RPMI-1640 + 20% FBS + 10% DMSO

  • Storage: Freeze at $3 - 5 \times 10^6\text{ cells/vial}$ using slow-freezing containers and store in liquid nitrogen (-196 degrees Celsius).
