一、 背景与来源说明 / Background & Origin
MUTU-I(常写作 MUTU 1 或 Mutu-1)是一种来源于人类地方性 Epstein-Barr 病毒(EBV)阳性伯基特淋巴瘤(Burkitt Lymphoma, BL)的 B 淋巴细胞株。
MUTU-I 与姊妹株 MUTU-III 同源,分离自同一名携带 $t(8;14)$ 染色体易位的男性患者活检组织。MUTU-I 属于严格的 EBV 潜伏期 I 型(Latency Type I)表型克隆。在该状态下,病毒基因组高度甲基化,仅表达 EBNA1 蛋白及 EBERs、ebv-miR-BART 等非编码 RNA,严格沉默 EBNA2、EBNA3s 及 LMP1/LMP2 等免疫原性蛋白。MUTU-I 是研究 EBV 潜伏期 I 型维持、染色质构象调控、病毒表观遗传学机制以及与 MUTU-III(潜伏期 III 型)对比研究潜伏期类型转换的金标准细胞模型。

三、 培养条件与传代指南 / Culture & Handling Protocols
1. 推荐培养基 / Complete Growth Medium
基础培养基:RPMI-1640
添加成分:10% 至 15% 灭活胎牛血清(FBS) + 1% 双抗(Penicillin-Streptomycin,可选)
培养环境:37 摄氏度,5% $CO_2$,高湿度培养箱
2. 复苏与传代操作 / Thawing & Subculture
复苏:将冻存管快速置于 37 摄氏度水浴中摇晃融化(约 1-2 分钟)。吸出细胞悬液加至含 8-10 mL 预热完整培养基的离心管中,125 x g 离心 5 分钟。弃上清,用预热培养基重悬,接种密度建议在 $0.4 - 0.6 \times 10^6\text{ cells/mL}$。
传代与维持:
维持细胞密度在 $0.3 - 0.6 \times 10^6\text{ cells/mL}$。
每 2 至 3 天按 1:2 或 1:3 比例补充新培养基或分瓶传代。
特殊注意:长期过度传代或高密度培养可能导致 MUTU-I 细胞自发漂移向潜伏期 III 型(高表达 LMP1/EBNA2),建议使用低代次细胞冻存保种,并定期通过 Western Blot 验证 LMP1/EBNA2 的阴性表达。
3. 冻存保存 / Cryopreservation
冻存液配方:70% RPMI-1640 + 20% FBS + 10% DMSO
冻存密度:$3 - 5 \times 10^6\text{ cells/vial}$,采用程序降温冻存并长期保存于液氮(-196 摄氏度)。

BioVector® MUTU-I Cell Line
1. Background and Origin
MUTU-I (also designated as MUTU 1 or Mutu-1) is a human B-cell line derived from an endemic Epstein-Barr virus (EBV)-positive Burkitt lymphoma (BL).
Isogenic to MUTU-III, MUTU-I was isolated from a biopsy of a male BL patient carrying the canonical $t(8;14)$ chromosomal translocation. MUTU-I strictly maintains the EBV Latency Type I phenotype. In this state, the viral genome is extensively methylated, selectively expressing only EBNA1 along with non-coding RNAs (EBERs and ebv-miR-BARTs), while silencing immunogenic viral proteins such as EBNA2, EBNA3s, and LMP1/LMP2. MUTU-I is the gold-standard cellular model for investigating EBV Type I latency maintenance, chromatin architecture, viral epigenetics, and latency phase transitions.

Recommended Growth Medium

Thawing and Subculture Protocol
Thawing: Rapidly thaw in a 37 degrees Celsius water bath. Transfer into 8–10 mL pre-warmed complete medium and centrifuge at 125 x g for 5 minutes. Resuspend in fresh medium and seed at $0.4 - 0.6 \times 10^6\text{ cells/mL}$.
Subculturing:
Maintain working cell density between $0.3 - 0.6 \times 10^6\text{ cells/mL}$.
Split cells 1:2 or 1:3 every 2–3 days.
Crucial Note: Extended passaging or overgrowth can induce spontaneous drift from Latency Type I toward Type III (upregulating LMP1 and EBNA2). Maintain low-passage stocks and regularly confirm LMP1/EBNA2 negativity via Western Blot.
Cryopreservation
Freezing Medium: 70% RPMI-1640 + 20% FBS + 10% DMSO
Storage: Freeze at $3 - 5 \times 10^6\text{ cells/vial}$ using slow-freezing containers and store in liquid nitrogen (-196 degrees Celsius).
